Commercial Scale Tissue Culture Propagation of a High Yielding Eucalyptus urophylla Clone in Gia Lai Province, Vietnam

Abstract
Introduction: The advantages of tissue-cultured seedlings compared to those produced seeds of by cuttings are that they are very disease-free, grow and develop well, are less prone to lodging in adverse weather conditions, and offer significantly higher economic efficiency. Eucalyptus trees are valuable for forest regeneration, providing raw materials for the pulp and paper industries and valuable medicinal properties.
Aim: A process for the rapid commercial scale propagation of high-yielding of primary clone of Eucalyptus urophylla has been developed.
Methods: Application of tissue culture techniques on internode cultivation. The study was conducted at: the Gia Lai Plant Breeding Research Center, Gia Lai province, Vietnam. Duaration of the experiment: January / 2012 – December / 2012. All the parameters were studied by using standard protocol.
Results: MS mineral medium was used as the basal medium for in vitro propagation of eucalyptus trees: full-MS + 0.1 mg/L IBA + 0.5 mg/L BA reach 5.80 shoots/cluster; induction of shoot cluster medium was MS + 0.1 mg/L IBA + 0.1 mg/L BA reach 5.03 shoots/cluster; multiple-shoot propagation medium: MS + 0.5 mg/L BA + 0.5 mg/L kinetin + 7% CW yielded high multiplication rate of 4.56 shoots/cluster and the other medium: MS + 0.1 mg/L BA + 0.25 mg/L IAA yielded the optimal multiplication rate of 5.30 shoots/cluster. Medium of 1/2MS + 0.25 mg/L NAA medium is suitable for in vitro rooting of eucalyptus plantlets.
Conclusion: A flush out system for eucalyptus production on commercial scale via internode cutting method was set up.
